This Massive Photonics DNA-PAINT Neuro-Imaging Kit is ready-to-use for the staining of the microtubule associated protein Tau using DNA-PAINT (DNA-Point Accumulation for Imaging in Nanoscale Topography). This localization based super-resolution microscopy method employs short, dye-labeled single stranded DNA that transiently binds its complementary sequences, which are attached to target-specific antibodies or nanobodies. Several Massive Photonics DNA-PAINT Neuro-Imaging Kits can be combined for multi-target staining and super-resolution imaging of various neuronal targets achieving visualization of cytoskeleton-associated and synaptic proteins with resolutions below 20 nm.

This kit provides ready-to-use binders for DNA-PAINT imaging of the neuronal microtubule associated protein Tau, which mainly localizes to axons. Also included are imager strands and buffers for sample preparation and imaging. This kit contains a monoclonal antibody recognizing Tau (cat. no. 314 011) that has been preincubated with single-domain antibodies (sdABs) from NanoTag conjugated to FAST-DNA-PAINT sequence F5. |

There are two major classes of heat stable microtubule associated proteins (MAPS): MAP 2 (280 kD), and tau (55-65 kD). Both protein classes are involved in the regulation of microtubule polymerization in cells. Tau is a neuronal protein that mainly localizes to axons. Hyperphosphorylated tau has been shown to be a major element of paired helical filaments in Alzheimer's disease.
