This Massive Photonics DNA-PAINT Neuro-Imaging Kit is ready-to-use for the staining of the scaffolding active zone protein Bassoon using DNA-PAINT (DNA-Point Accumulation for Imaging in Nanoscale Topography). This localization based super-resolution microscopy method employs short, dye-labeled single stranded DNA that transiently binds its complementary sequences, which are attached to target-specific antibodies or nanobodies. Several Massive Photonics DNA-PAINT Neuro-Imaging Kits can be combined for multi-target staining and super-resolution imaging of various neuronal targets achieving visualization of cytoskeleton-associated and synaptic proteins with resolutions below 20 nm.

DNA-PAINT: yes gallery  

In antibody-based DNA-PAINT (Point Accumulation in Nanoscale Topography), a short oligomeric docking strand is coupled to a specific antibody. The transient association of the fluorophore to the antibody is mediated by the pairing of a short fluorescently labeled complementary imager DNA strand. DNA-PAINT allows super-resolution imaging and the imaging of a huge number of antibodies on the same biological sample in a single multiplex experiment. This kit provides ready-to-use binders for DNA-PAINT imaging of the scaffolding protein Bassoon, which is essential for the organization of the presynaptic active zone and the regulation of neurotransmitter release. Also included are imager strands and buffers for sample preparation and imaging.

This kit contains a monoclonal antibody recognizing Bassoon (cat. no. 141 111) that has been preincubated with single-domain antibodies (sdABs) from NanoTag conjugated to FAST-DNA-PAINT sequence F4.

Background

Bassoon is a large protein which consists of an N-terminal Zn2+ finger and several piccolo-bassoon homology domains (PBH-domains). It is generally found together with piccolo, a related huge multi-domain protein of the CAZ (cytoskeletal matrix assembled at active zones).
Bassoon was suggested to be a scaffolding element of the presynapse but deletion experiments in mice have shown that bassoon is also involved in synaptic vesicle cycling. Probably bassoon interacts with other protein factors via its Zn2+ domain but the potential partners have not been determined yet.
